NR8383 (正常大鼠,1983年8月3日)來源於肺灌洗時的正常大鼠肺泡巨噬細胞。 細胞在gerbil肺細胞連續培養液存在下培養了大約8-9個月。 隨後,不再需要外源生長因子。通過有限稀釋法從單個細胞克隆並亞克隆NR8383細胞,並三次用軟瓊脂亞克隆。 細胞表現出巨噬細胞的特性,吞噬酵母多糖和銅綠,非特異性脂酶活性,Fc受體,氧化降解;分泌IL-1, TNF beta和IL-6,可重複地響應外源生長因子。 NR8383細胞響應博萊黴素,分泌TGF beta前體。在博萊黴素刺激下,TGF beta mRNA表達也上升。 細胞對內**敏感。 1-10 鈉克/毫升的LPS水平抑製增生達50%。 即使達到0.001毫克/毫升的水平,LPS抑製還是無毒且在後續過程中可逆的。 NR8383細胞株提供了高響應的肺泡巨噬細胞的均一來源,可以用於體外研究巨響細胞相關活性。
