Cytogenetic Analysis: SNU-1 is a near-diploid cell line with a modal chromosome number of 47; double minute chromosomes were found in 28% of the cells. They have a reported colony-forming efficiency of 1.9% in semisolid medium and a population doubling time of 26 hours. The cells grow as floating aggregates of round cell clusters. This is a hyperdiploid human cell line with the modal chromosome number of 47 occurring in 94% of cells. Higher ploidies occurred at 42%, which is very high. The karyotype of 47,XY,t(1;4) (q24;q33), +M1 is very uniform and stable. M1 is an F- sized small metacentric chromosome.

Comments: SNU-1 was derived in 1984 by J. Park and associates from a poorly differentiated primary carcinoma of the stomach, taken prior to cytotoxic therapy. The cells are L-dopa decarboxylase (DDC) negative. SNU-1 cells were positive for VIP receptors but lacked gastrin receptors. No evidence of amplification or rearrangements was noted in the N-myc, L-myc, myb and EGF receptor genes. The cell line expressed levels of c-myc and c-erb-B-2 RNA that were comparable to other cell lines. There was no expression of the following genes: N-myc, L-myc, c-cis, IGF-2, or gastrin releasing peptide.
Propagation: ATCC complete growth medium: The base medium for this cell line is ATCC-formulated RPMI-1640 Medium, Catalog No. 30-2001. To make the complete growth medium, add the following components to the base medium: fetal bovine serum to a final concentration of 10%.
