 The ImmEdge Hydrophobic Pen from Vector Laboratories is a very useful tool for those performing immunohistochemistry utilizing paraffin-embedded tissue sections. Previously we were using plastic coverslips, which would sometimes introduce unwanted air bubbles and occasionally interfere with the tissue section itself. In our hands, the ImmEdge pen is simple to use and one of its main advantages is that significantly less antibody need be applied. In general, we use 50 – 100µl of antibody per section (depending on section size) when using the ImmEdge pen compared to 100 – 200µl of antibody when using the plastic coverslip method.
The basicideaoftheImmEdge pen lies in the hydrophobic characteristicsofitscontents.This enables the user to circumscribe aroundthetissuesection in adefined manner, forming a hydrophobicbarrier.Uponapplication ofthe antibody the solution remainsprecisely onthetissue sectionwithin the boundariesofthehydrophobicbarrier.
Thepensaresuppliedin packs of two and when received are in a drystate.Tostart theflow of the hydrophobic solution, the tip of thepenisdepressedseveral times until the tip is completelysaturatedwithsolution.It is advisable not to be too vigorous inthisprocedureas this canlead to subsequent pen leakage. When usingthepen,care should betaken that the hydrophobic barrier iscompletelydrybefore applyingthe antibody solution and it should beensuredthatno gaps arepresent. If the flow of solutionbecomesinadequate,the tip of thepen can be depressed toincreasetheflow.
Thehydrophobicbarrier solution is insoluble inmostalcohols andacetone so onceapplied it is often unnecessarytoreapply atsubsequent steps ofstandard immunostaining protocols.Forthosedehydrating sectionsfollowing processing of thesectionsforvisualization via lightmicroscopy the barrierisconvenientlyremoved upon immersion ineither xylene orcommonly-usedxylenesubstitutes. In our extensiveexperience thepresence of thebarrierdoes not significantlyinterfere withroutineimmunofluorescenceprotocols when usingaqueous mountingmedium suchas VectorLaboratoriesVectaShield.
